ABSTRACT

Condensable” or “packable” composites (Surefil) were introduced as an alternative to dental amalgam. However, concerns have been raised related to the ability of these stiffer materials to adequately adapt to internal surfaces. To offset this problem, low viscosity fiowable liners walls are recommended under packable composites. This study aimed at evaluating the effect of SDR and Ketac N100 liners on marginal sealing ability of High Density Surefil posterior packable composite. Methodology: Sixty permanent extracted molars were mounted in a modelling wax and box only Class II cavities were prepared on the mesial side with gingival seat 1mm above CEJ. Teeth were divided into 3 groups: Group (i) Surefil with SDR; Group (ii) Surefil with Ketac N 100; and Group (iii) Surefil without liner. After restoration, the teeth were immersed in Methylene blue dye, sectioned mesiodistally and then evaluated for leakage under Stereomicroscope. Results: None of the groups were free of microleakage. Surefil without liner (1.2±0.696) exhibited maximum microleakage followed by Ketac N100 group (0.9±0.718) and SDR group (0.8±0.523) showed least microleakage. Conclusion: Posterior packable composites should be lined by a resin based liner at the gingival margin to allow better seal

ABSTRACT

OBJECTIVES: This study aimed to compare the shear bond strength (SBS) of a self-adhering flowable composite (Dyad Flow) and a bulk-fill flowable composite (Smart Dentin Replacement [SDR]) to several pulp-capping materials, including MTA Plus, Dycal, Biodentine, and TheraCal.MATERIALS AND METHODS: Eighty acrylic blocks with 2-mm-deep central holes that were 4 mm in diameter were prepared and divided into 2 groups (n = 40 each) according to the composite used (Dyad Flow or SDR). They were further divided into 4 sub-groups (n = 10 each) according to the pulp-capping agent used. SBS was tested using a universal testing machine at a crosshead speed of 1 mm/min. Data were analyzed using 2-way analysis of variance. A p value of < 0.05 was considered to indicate statistical significance.RESULTS: A statistically significant difference (p = 0.040) was found between Dyad Flow and SDR in terms of bond strength to MTA Plus, Dycal, Biodentine, and TheraCal.CONCLUSIONS: Among the 8 sub-groups, the combination of TheraCal and SDR exhibited the highest SBS.

ABSTRACT

Introduction: Even with the advances and innovations inrestorative dentistry, microleakage is regarded one of the mainproblems in this area. Objective: To evaluate in vitro marginal microleakage ofBiodentine, Fuji II LC and SDR at the cervical level and at theinterface these materials with Tetric N Ceram Bulk fillcomposite and also to note the marginal microleakage whenthese materials were used as bulk fill materials.Materials and Methods: Cavities, standardized on the mesialand distal surfaces, were prepared in thirty molars andrandomly assigned to two groups (n = 30), according to themode of restoration done.Goup1 (control) in which testmaterials were used as bulk fill. These were further subdividedinto subgroups according to the material. SG1: Biodentine(SEPTODONT); SG2: Fuji II LC (GC); SG3: SDR (Dentsply).Group2 (experimental) in which test materials used as dentinsubstituent in class II sandwich technique followed byrestoration with Tetric N Ceram (Ivoclar vivadent). These werealso divided into subgroups, SG4: Biodentine + Tetric NCeram; SG5: Fuji II LC + Tetric N Ceram; SG6: SDR + Tetric NCeram. After storage for 24 hours in an incubator (37 °C), thesamples were submitted to the thermocycling test (500 cycles:5 °C/55 °C). They were later waterproofed, immersed in 1%methylene blue solution and sectioned in the mesialdistal direction for evaluation under stereomicroscope at 30xmagnification (Nikon SMZ 1500 Zoom Stereomicroscope)Scores from 0 to 3 using the ISO microleakage scoring system(ISO/TS 11405:2003). The ANOVA Test and Post- hoc Test, with a significance level of 5%, were used for statisticalanalysis. Conclusion: The study concluded that SDR (SD 2.38) showedthe least amount of microleakage both at the occlusal and atthe cervical levels in open sandwich restorations and alsowhen used as bulk fill materials as compared with Biodentine(SD 1.73) and Fuji II LC (SD 1.00).

ABSTRACT

Fusidic acid is the only fusidane-type antibiotic that has been clinically used. However, biosynthesis of this important molecule in fungi is poorly understood. We have recently elucidated the biosynthesis of fusidane-type antibiotic helvolic acid, which provides us with clues to identify a possible gene cluster for fusidic acid ( cluster). This gene cluster consists of eight genes, among which six are conserved in the helvolic acid gene cluster except and . Introduction of the two genes into the NSAR1 expressing the conserved six genes led to the production of fusidic acid. A stepwise introduction of and revealed that the two genes worked independently without a strict reaction order. Notably, we identified two short-chain dehydrogenase/reductase genes and in the cluster, which showed converse stereoselectivity in 3-ketoreduction. This is the first report on the biosynthesis and heterologous expression of fusidic acid.

ABSTRACT

ABSTRACT

ABSTRACTThe serine-aspartate repeat proteins (Sdr) are members of a family of surface proteins and contribute to the pathogenicity of Staphylococcus aureus. Among 288 S. aureus isolates including 158 and 130 associated with skin and soft tissue infections and bloodstream infection, respectively; 275 (95.5%) were positive for at least one of threesdr genes tested. The positivity rates for sdrC, sdrD, and sdrE among S. aureusisolates were 87.8% (253/288), 63.9% (184/288), and 68.1% (196/288), respectively. 224 (77.8%) of 288 isolates were concomitantly positive for two or three sdr genes. There was an association between carriage ofsdrE and methicillin-resistant S. aureus(MRSA) isolates, while the carriage rates of sdrC andsdrD in MRSA isolates were similar to those in methicillin-sensitive S. aureus (MSSA) isolates. The prevalence of co-existence of sdrC and sdrE among MRSA isolates was significantly higher than that among MSSA isolates (p < 0.05). All ST1, ST5, ST7, and ST25 isolates were positive for sdrD. While all ST121 and ST398 isolates were negative for sdrD. All ST59 and ST88 isolates were positive forsdrE. All ST1 isolates were concomitantly positive forsdrC and sdrD. Concomitant carriage ofsdrC, sdrD, and sdrE was found among all ST5, 75.0% (9/12) of ST1, 69.2% (9/13) of ST6, 78.6% (11/14) of ST25, and 90.9% (20/22) of ST88 isolates. sdrD was linked to CC5, CC7 and CC88 isolates, especially CC88 isolates. There was a strong association between the presence of sdrE and CC59, CC88, and CC5 isolates. A significant correlation between concomitant carriage of sdrC, sdrD, and sdrE and CC88 isolates was found.sdrC-positive, sdrD-positive andsdrE-negative gene profile was significantly associated with CC7 clone. There was an association between sdrC-positive,sdrD-negative, and sdrE-positive gene profile and CC59 isolates. A correlation between sdrC-positive,sdrD-negative, and sdrE-negative gene profile and CC121 clone was found. More CC59 isolates carriedsdrC-negative, sdrD-negative, andsdrE-positive gene profile relative to other four CCs isolates. All ST1 and ST5, 95.2% (20/21) of ST188 and 95.2% (20/21) of ST630 isolates were positive for sdrC. Taken together, our investigation indicated that different S. aureus lineages were associated with specific patterns of carriage of sdr genes.

ABSTRACT

Bone sialoprotein-binding protein (Bbp), a MSCRAMMs (Microbial Surface Components Recognizing Adhesive Matrix Molecules) family protein expressed on the surface of Staphylococcus aureus (S. aureus), mediates adherence to fibrinogen α (Fg α), a component in the extracellular matrix of the host cell and is important for infection and pathogenesis. In this study, we solved the crystal structures of apo-Bbp(273-598) and Bbp(273-598)-Fg α(561-575) complex at a resolution of 2.03 Å and 1.45 Å, respectively. Apo-Bbp(273-598) contained the ligand binding region N2 and N3 domains, both of which followed a DE variant IgG fold characterized by an additional D1 strand in N2 domain and D1' and D2' strands in N3 domain. The peptide mapped to the Fg α(561-575) bond to Bbp(273-598) on the open groove between the N2 and N3 domains. Strikingly, the disordered C-terminus in the apo-form reorganized into a highly-ordered loop and a β-strand G'' covering the ligand upon ligand binding. Bbp(Ala298-Gly301) in the N2 domain of the Bbp(273-598)-Fg α(561-575) complex, which is a loop in the apo-form, formed a short α-helix to interact tightly with the peptide. In addition, Bbp(Ser547-Gln561) in the N3 domain moved toward the binding groove to make contact directly with the peptide, while Bbp(Asp338-Gly355) and Bbp(Thr365-Tyr387) in N2 domain shifted their configurations to stabilize the reorganized C-terminus mainly through strong hydrogen bonds. Altogether, our results revealed the molecular basis for Bbp-ligand interaction and advanced our understanding of S. aureus infection process.

ABSTRACT

To estimate the effectiveness and efficiency of pulmonary surfactants used in infants with respiratory distress syndrome at the Colombian Cardiovascular Foundation, we designed an observational, descriptive study. We analyzed the medical records of newborns hospitalized in this institution between January 2009 and April 2011 and calculated the effectiveness and efficiency indicators for each surfactant. After analyzing aggregate data, we found that the most efficient and effective medication for these patients was Survanta. It requires fewer doses per treatment, is more economical, and generates fewer days in the intensive care unit and fewer days of mechanical ventilation. In some cases, however, Infasurf was more effective than Survanta, caused less extubation failure in infants with low birth weight and fewer complications during treatment...
